Olive oil polyphenols, which possess cytoprotective activities like anti-oxidant and anti-inflammatory effects, could modulate osteoblast functions. The aim of this study is to elucidate the effects underlying mechanisms hydroxytyrosol oleuropein on tumor necrosis factor-α (TNF-α)-induced macrophage colony-stimulating factor (M-CSF) interleukin-6 (IL-6) synthesis in osteoblasts.Osteoblast-like MC3T3-E1 cells were pretreated with hydroxytyrosol, oleuropein, deguelin, PD98059 or wedelolactone, then stimulated by TNF-α. levels M-CSF IL-6 conditioned medium determined ELISA. mRNA expression real-time RT-PCR. phosphorylation Akt, p44/p42 mitogen-activated protein (MAP) kinase NF-κB cell lysates Western blot analysis.Hydroxytyrosol attenuated TNF-α-stimulated release. Deguelin, an inhibitor significantly suppressed release, failed be affected MEK1/2 IκB wedelolactone.
