ABSTRACT
Dendrobium mixture (DM) is a patent Chinese herbal formulation consisting of Dendrobii Caulis, Astragali Radix, Rehmanniae Radix as the main ingredients. DM has been shown to alleviate diabetic related symptoms attributed its anti-hyperglycaemic and anti-inflammatory activities. However, effect on induced cognitive dysfunction not investigated. This study aims investigate in improving impairment associated mechanisms. Our confirmed showed capacity restore memory function high fat/high glucose streptozotocin-induced rats. The neuroprotective was manifested improved learning behaviours, restored blood-brain barrier tight junction, enhanced expressions neuronal survival biomarkers. protected colon effectively lowered circulated proinflammatory mediators including tumour necrosis factor-α, interleukin-6 lipopolysaccharides. In gut microbiota, corrected increase abundance Firmicutes, ratio Firmicutes/Bacteroidetes, decrease Bacteroidetes It also reversed Lactobacillus, Ruminococcus Allobaculum genera. Short chain fatty acids, isobutyric acid ethylmethylacetic acid, were negatively significantly correlated Allobaculum. Isovaleric positively with which all contributing improvement level, systemic inflammation results demonstrated potential promising therapeutic agent treating underlying mechanism may be regulating microbiota.
