ABSTRACT
Platelets are versatile anucleate cells involved in thrombosis as well inflammation. Stachydrine (STA), a major bioactive compound extracted from Motherwort, has multiple pharmacological properties. Nevertheless, the significance of STA platelet regulation and whether could ameliorate platelet-mediated thrombo-inflammation still remain elusive.Human platelets were used to assess regulatory effects on activation interactions with neutrophils vitro. FeCl3 injury-induced carotid/mesenteric collagen/epinephrine-induced pulmonary thromboembolism model explore regulate vivo. Furthermore, cecal ligation puncture-induced sepsis was employed investigate role thrombo-inflammatory diseases.STA markedly suppressed represented by aggregation, secretion, αIIbβ3-mediated signaling events calcium mobilization, etc. inhibiting agonists-induced potentiating cGMP-dependent inhibitory signaling. Mice receiving STA-treated less susceptible In addition, decreased platelet-neutrophil including aggregates neutrophil extracellular traps, alleviative sepsis-induced multiorgan damage observed due STA-mediated inhibition. This study suggested potential therapeutic thrombotic disorders.
