ABSTRACT
Liver fibrosis is a chronic and progressive disease with complex pathogenesis related to bile acids (BAs) gut microbiota. Forsythiaside A (FTA), isolated from the traditional Chinese medicine Forsythiae Fructus (Lian Qiao), natural hepatoprotective agent. The purpose of this study was investigate protective effect FTA on carbon tetrachloride (CCl4)-induced liver in mice. induced mice by intraperitoneal injection 2 mL/kg CCl4 three times week for 4 weeks. attenuated CCl4-induced mice, which proved results Masson Sirius red staining, hydroxyproline, hyaluronic acid, laminin, type III procollagen, IV collagen assays. inhibited hepatic stellate cell activation, reduced inflammation oxidative stress treated CCl4. What's more, ameliorated dysbiosis, maintained intestinal barrier function, increased production short-chain fatty (SCFAs), improved endotoxemia, as manifested decreased serum lipopolysaccharide levels expression ileal tight junction proteins. Besides, can modulate genes acid metabolism alter distribution fecal BAs fibrotic In word, improve inhibiting stress, regulating microbiota BA metabolism, increasing content SCFAs. provided an important reference mechanisms products prevent fibrosis.
