ABSTRACT
Caffeic acid phenethyl ester (CAPE), a bioactive component of propolis, has beneficial effects on cancer prevention. Growth differentiation factor 15 (GDF15) is an antitumor gene bladder cancer. Therefore, this study investigated the anti-cancer effect CAPE carcinoma cells and related mechanisms. The expressions GDF15, N-myc downstream-regulated 1 (NDRG1), maspin, activations extracellular signal regulated kinase (ERK), c-jun Nterminal (JNK), p38, 50 adenosine monophosphate-activated protein (AMPK) α1/2 in human after transfection or knockdown were determined by immunoblot, real-time reverse transcriptase-polymerase chain reaction (RT-qPCR), reporter assays. assays 5-ethynyl-2'-deoxyuridine (EdU), CyQUANT cell proliferation, Matrigel invasion, xenograft animal used to assess tumorigenesis. GDF15 expression epithelial was negatively correlated with neoplasia vitro. Also, exhibits fibroblasts smooth muscle cells. CAPE-induced NDRG1 maspin decreased proliferation invasion GDF15-dependent manner suggesting attenuated tumor growth vivo. increased phosphorylation ERK, JNK, AMPKα1/2 modulate expressions. Pretreatments p38 inhibitors partially inhibited inductions NDRG1, maspin. Knockdown Our findings indicate that promising agent for anti-tumor via upregulation GDF15.
