ABSTRACT
In the present study, we evaluated an autologous bone graft substitute (ABGS) composed of recombinant human BMP6 (rhBMP6) dispersed within blood coagulum (ABC) used as a physiological carrier for new formation in spine fusion sheep models. The application ABGS included cervical cage use anterior lumbar interbody (ALIF), while posterolateral (PLF) model allograft devitalized particles (ALLO) were applied with and without instrumentation. ALIF model, (rhBMP6/ABC/cage) implants fused significantly when placed between L4-L5 vertebrae compared to control (ABC/cage) which appears have fibrocartilaginous gap, examined by histology micro CT analysis at 16 weeks following surgery. PLF or ALLO showed complete ectopically gutter bilaterally two decorticated transverse processes success rate 88% instrumentation 80% instrumentation; however volume was 50% lower group than without, histology, radiographs, analyses biomechanical testing 27 newly formed uniform resulting biomechanically competent histologically qualified optimum dose range 100 μg rhBMP6 per mL ABC, that contained ALLO, mineralized substituted remodeling via creeping substitution. These findings demonstrate first time (rhBMP6/ABC) induced robust successful models PLF, can serve preferred native induce low doses achieve spinal fusion.
