ABSTRACT
The DNA tumor virus Simian 40 (SV40) is a model system for studying eukaryotic replication. SV40 large antigen (LTag) the initiator/helicase that essential genome LTag recognizes and assembles at viral replication origin. We determined structure of two multidomain subunits bound to origin DNA. reveals binding domains (OBDs) Zn AAA+ are involved in recognition assembly. Notably, OBDs recognize an unexpected manner. histidine residues insert into narrow minor groove region with enhanced negative electrostatic potential. Computational analysis indicates this intrinsically narrow, demonstrating role shape readout recognition. Our results provide important insights assembly suggest contacts serve as mechanism readout.
