ABSTRACT
Hypertrophic scars (HS) are a variety of skin tissue fibrosis disease that occurs in human skin, the effective therapeutic method which is still inaccessible up to now. As bioactive constituent well-known medical plant, Salvia miltiorrhiza (Danshen Chinese), tanshinone IIA (TSA) reported inhibit cell proliferation HS. Therefore, aim this study was prepare TSA self-soluble microneedles strengthen its dermal retention and break through difficulty significantly thickening epidermal connective stratum corneum at HS site. The possible mechanism action suppressing studied using fibroblasts (HSF).Tanshinone self-dissolving (TSA-MN) prepared negative mold casting method. prescription process microneedle optimized by Box-Behnken effect surface Different media were selected investigate ability transdermal absorption vitro release. Furthermore, according Cell Counting Kit-8 (CCK8) as well Western blot method, TSA-MN on biological characteristics HSF investigated. With remarkable slow release retention, properties better than both ordinary dosage forms. Its confirmed essential decrease motility during migration vitro, plays significant role down-regulating secretion transforming growth factor-β1 (TGF-β1) increasing expression level Smad7.The helpful solving problem hypertrophic scars, with stable after optimization.
