The genome of the rice variety LTH provides insight into its universal susceptibility mechanism to worldwide rice blast fungal strains

ABSTRACT
The widely used rice variety Lijiangxintuanheigu (LTH) shows a universal susceptibility to thousands of Magnaporthe oryzae isolates, the causal agent devastating blast, making LTH an ideal line in resistance (R) gene cloning. However, underlying genetic mechanism has not been fully revealed because lack high-quality genome. Here, we took genomic approach together with experimental assays investigate LTH's blast. Using Nanopore long reads, assembled chromosome-level Millions variants were detected by comparing 10 other varieties, which large-effect could affect plant immunity. Gene family analyses show that number R genes and leucine-rich repeat receptor-like protein kinase (LRR-RLK)-encoding decrease significantly LTH. Rice blast called Pi are either absent or disrupted variations. Additionally, residual likely under weak pathogen selection pressure, defense-related weakly induced In contrast, pattern-triggered immunity (PTI) is normal, as demonstrated assays. Therefore, conclude effector-trigger (ETI)-mediated primarily but PTI results attenuated ETI may be also associated reduced numbers LRR-RLKs, minimally functional genes. Finally, demonstrate use genome rapid cloning Piak from resistant variety.
