Deciphering m6A dynamics at a single-base level during planarian anterior-posterior axis specification

ABSTRACT
BackgroundThe establishment of the anterior-posterior (A-P) axis is a crucial step during tissue repair and regeneration. Despite association reported recently N6-methyladenosine (m6A) with regeneration, mechanism underlying regulation m6A in A-P specification regeneration remains unknown. Herein, we deciphered landscape at single-base resolution multiple time points constructed de novo transcriptome assembly Dugesia japonica planarian.ResultsImmunofluorescence staining comparative analysis revealed that widespread across planarian dynamically regulated along axis, exhibiting strong spatiotemporal feature. The resulting datasets m6A-modified genes identified 80 anterior-specific 13 posterior-specific genes, respectively. In addition, showed YTHDC1 serves as primary reader to be involved m6A-mediated planarian.ConclusionsOur study provides an RNA epigenetic explanation for planarian.
