ABSTRACT
Highlights•Transgenic skinbow zebrafish barcodes skin superficial epithelial cells (SECs)•Hundreds of SECs are simultaneously monitored in vivo over cell surface lifetimes•SEC behaviors quantified during regeneration wounds or amputation injuries•Identification and quantification ROS effects on SEC regenerationSummaryCurrent fate mapping imaging platforms limited their ability to capture dynamic cells. To deconstruct regenerating adult tissue at single-cell resolution, we created a multicolor system, skinbow, that the (SEC) population with dozens distinguishable tags. With image analysis directly segment track hundreds entire lifetimes, readily orchestration emergence, growth, repositioning, loss under homeostatic conditions after exfoliation appendage amputation. We employed skinbow-based conjunction live reporter stem cycle activity as an instrument evaluate reactive oxygen species behavior regeneration. Our findings introduce platform for large-scale, quantitative reveal unanticipated collective dynamism size, mobility, interactions.Graphical abstract
