ABSTRACT
Polybrominated diphenyl ethers (PBDE-47), a kind of lipophilic persistent organic pollutants (POPs) brominated flame retardant, has been widely used in various consumer products. However, the toxicity PBDE-47 on human immune system not well elucidated. Neutrophil extracellular traps (NETs) contribute to innate responses, and release NETs is recognized as most important part killing mechanism. The aim this study was investigate effect its possible molecular mechanism, intervention curcumin (Cur). In study, formation PBDE-47-induced observed by fluorescence microscopy scanning electron microscopy, also quantitatively detected DNA dye SYTOX green. addition, we Cur Nrf2 inhibitor ML385 explore role reactive oxygen species (ROS), signal regulated kinase (ERK) p38 signaling pathway reticular formation. We demonstrated that could significantly induce NETs, mechanism might be related ROS burst. reduced inhibited interfering with Nrf2. conclusion, revealed hindered via Nrf2-associated inhibition, which enriched cytotoxicity PBDE-47, provided new clue for development an antagonist PBDE-47-related injury.
