ABSTRACT
Vero cells have been used successfully in Toxoplasma gondii maintenance. Medium supplementation for culture with fetal bovine serum is necessary cellular growth. However, these cultures presents disadvantages, such as the potential to induce hypersensitivity, variability of batches, possible presence contaminants, and high cost good quality serum. Culture media formulated without any animal derived components, designed serum-free growth cell lines different virus replication. The advantages protozoan parasite line using medium remain poorly studied. Thus, this study was determine whether T. tachyzoites grown medium, after many passages, are able maintain same antigenic proprieties those maintained experimental mice. standardization vitro tachyzoite production performed establishing optimal initial concentration confluent monolayer formation, which 1 × 106 inoculum. total formatted 96 h best amount harvested 2.1×107 inoculum 1.5 7 days post-infection. infectivity released from evaluated groups Swiss mice infected cell-culture tachyzoites. concentrations were similar collected other data both vivo experiments showed that at least 30 parasites vivo. Another question know several continued immunogenic progressive loss could occur. nucleotide sequences studied between mean no change their viability lysate antigen. antigen by has clear ethical cost-saving advantages. Moreover, use human or lines, produced especially production.
