ABSTRACT
ObjectiveClear cell renal carcinoma (ccRCC) is the most common subtype among carcinomas and has worst prognosis, originating from tubular epithelial cells. Toll-like receptor 4 (TLR4) plays a crucial role in ccRCC proliferation, infiltration, metastasis. The aim of this study was to construct prognostic scoring model for based on TLR4 expression heterogeneity explore its association with immune thereby providing insights treatment evaluation ccRCC.MethodsUsing R software, differential analysis conducted normal samples samples, conjunction KEGG database, correlation clear pathway (hsa05211) carried out. We observed TCGA-KIRC cohort identified related genes (TRGs). Based levels TRGs, consensus clustering employed identify TLR4-related subtypes, further heatmaps, principal component, single-sample gene set enrichment analyses were conducted. Overlapping (ODEGs) between subtypes analysed, combined survival data, univariate Cox regression, LASSO, multivariate regression used establish risk ccRCC. This subsequently evaluated through ROC analysis, factor independent intergroup analysis. ssGSEA ccRCC, performance predicting patient prognosis using box plots oncoPredict software package.ResultsIn cohort, notably elevated compared correlating improved high-expression group. distinct categorized into three varied outcomes. A overlapping established, showing significant associations infiltration key checkpoints (PD-1, PD-L1, CTLA4). Additionally, drug sensitivity differences groups.ConclusionIn exhibited heterogeneity. Through we that primary cells across are myeloid-derived suppressor cells, central memory CD4 T regulatory Furthermore, successfully constructed composed 17 genes. provides valuable references prediction patients.
