ABSTRACT
BackgroundColorectal cancer (CRC) is one of the most common malignancies, and pyroptosis exerts an immunoregulatory role in CRC. Although location primary tumor a prognostic factor for patients with CRC, mechanisms left- right-sided CRC remain unclear.MethodsExpression clinical data were collected from The Cancer Genome Atlas Gene Expression Omnibus databases. Differences characteristics, immune cell infiltration, somatic mutations between then compared. After screening differentially expressed genes, Pearson correlation analysis was performed to select pyroptosis-related followed by gene set enrichment analysis. Univariate multivariate Cox regression analyses used construct validate model nomogram predicting prognosis. Collected samples subjected quantitative reverse transcription-quantitative PCR (RT-qPCR) expression key genes.ResultsLeft- exhibited significant differences features infiltration. Five signatures identified among 134 genes risk score-based model, adverse outcomes high-risk further verified using external cohort. A also generated based on three independent factors predict survival probabilities, while calibration curves confirmed consistency predicted actual survival. Experiment differential five CRC.ConclusionThe may be potential biomarkers prognosis help improve
