Integrated analysis of whole blood oxylipin and cytokine responses after bacterial, viral, and T cell stimulation reveals new immune networks

ABSTRACT
Oxylipins are major immunomodulating mediators, yet studies of inflammation focus mainly on cytokines. Here, using a standardized whole-blood stimulation system, we characterized the oxylipin-driven inflammatory responses to various stimuli and their relationships with cytokine responses. We performed pilot study in 25 healthy individuals 6 different stimuli: 2 bacterial (LPS live BCG), viral (vaccine-grade poly I:C H1N1 attenuated influenza), an enterotoxin superantigen Null control. All induced strong production oxylipins but most importantly, bacterial, viral, T cell immune show distinct oxylipin signatures. Integration for each condition revealed new networks improving our understanding regulation. Finally, oxylipin-cytokine were compared patients active tuberculosis or latent infection. This BCG not LPS highlighting regulatory pathways further investigations.
