Molecular identification, phylogeny and antifungal susceptibilities of dematiaceous fungi isolated from human keratomycosis

ABSTRACT
To investigate the dematiaceous fungal profile of patients with ocular mycoses attending a tertiary eye care hospital in Coimbatore, India METHODS: The identification fungus based on their morphology, genotypes, and measurement minimum inhibitory concentrations (MICs) using microdilution method routinely used antifungal drugs were all compared.A total 148 fungi isolated during study period 27 months. Isolates confirmed as Curvularia spp. (n = 98), Exserohilum 32), Alternaria 14), Exophiala 2), Cladosporium sp. 1) Aureobasidium 1). Out 50 well grown isolates characterized genotypically amplification sequencing ITS region ribosomal RNA gene cluster subsequent BLAST analysis, lunata 24), C. aeria 1), spicifera 8), hawaiiensis maydis papendorfii geniculata 3), tetramera 2) Exs. rostratum 7) identified. In vitro susceptibilities most tested showed that voriconazole had MIC50 0.25 μg ml-1, while amphotericin B ml-1 for spp.Voriconazole proved to be effective drug against pigmented filamentous fungi, followed by B, itraconazole econazole.
