ABSTRACT
Daphnetin (DAP) is a traditional Chinese drug usually used to treat cardiovascular diseases. Studies have confirmed the anti-inflammatory, antioxidant, anti-bacterial and insecticidal, anti-tumor neuro-protective effects of DAP. However, its anti-arthritic potential remains unexplored. The aim this study investigate in vitro vivo chondroprotective DAP.The effect DAP on primary rabbit chondrocytes was examined using recombinant human IL-1β for 24 h. For studies, rabbits were randomly divided into groups: normal control group osteoarthritis (OA) groups. OA groups received three different doses 4 or 8 weeks. assessed histopathological examinations, qRT-PCR, western blotting immunohistochemical analysis.Both results indicate that exerts protective against chondrocytes. In vitro, inhibits expression IL-6, IL-12, MMP-3, MMP-9 MMP-13, induced by chondrocytes, stimulates production IL-10. inhibitory MMPs partially regulated inhibition PI3K/AKT, MAPK NF-κB signaling pathways. may be attributed suppression inflammatory factor secretion, chondrocyte apoptosis observed decrease pro-apoptotic Caspase-3 BAX, activation anti-apoptotic BCL-2.DAP has broad range prospects treatment OA, which provides novel therapeutic strategy OA.
