Identification of distinct immune infiltration and potential biomarkers in patients with liver ischemia-reperfusion injury

ABSTRACT
To identify alterations of specific gene expression, immune infiltration components, and potential biomarkers in liver ischemia-reperfusion injury (IRI) following transplantation (LT).GSE23649 GSE151648 datasets were obtained from the Gene Expression Omnibus (GEO) database. determine differentially expressed genes (DEGs), we utilized R package "limma". We also different cells through single-sample gene-set enrichment analysis (ssGSEA). Furthermore, LASSO logistic regression to select feature Spearman's rank correlation between these infiltrating cells. Finally, significance was confirmed using a mouse model hepatic IRI.A total 17 DEGs acquired, most which associated with inflammation, apoptosis, cell proliferation, disorders, cellular response. 28 types determined ssGSEA. 5 (ADM, KLF6, SERPINE1, SLC20A1, HBB) screened analysis, but HBB ultimately excluded due lack statistical dataset. These 4 predominantly related 15 significantly distinctive control IRI groups verified.We unveiled that macrophages, dendritic (DCs), neutrophils, CD4 T cells, other infiltrated occurred after LT. Moreover, identified ADM, SLC20A1 as biological underlying post-transplant, may improve diagnosis prognosis this condition.
