ABSTRACT
The research article describing the discovery of ribosomal frameshifting in bacterial CopA gene also reported occurrence expression human ortholog ATP7B based on assays using dual luciferase reporters. An examination publicly available ribosome profiling data and phylogenetic analysis proposed site cast doubt validity this claim prompted us to reexamine evidence. We observed similar apparent efficiencies as original authors same type vector that synthesizes both luciferases a single polyprotein. However, we noticed anomalously low absolute activities from N-terminal reporter suggests interference activity or levels by test cassette. When tested cassette more recently developed system which reporters are released without being included polyprotein, no was detected above background levels.
