Bcl-2 supports survival and metabolic fitness of quiescent tissue-resident ILC3

ABSTRACT
Group 3 innate lymphoid cells (ILC3) are potent effector with critical roles in enforcing immunity, barrier integrity and tissue homeostasis along the gastrointestinal tract. ILC3 considered primarily tissue-resident cells, seeding tract during embryonic stages early life. However, mechanisms through which mature maintained within adult tissues poorly understood. Here, we report that tissue-inducer-like (LTi-like) exhibit minimal turnover healthy intestinal tract, persist for extended periods of time, display a quiescent phenotype. Strikingly, enteric bacterial infection LTi-like also negligible hematopoietic replenishment remain non-proliferative, despite robustly producing cytokines. Survival was found to be dependent upon balance between metabolic activity required drive function anti-apoptotic programs. Notably, pro-survival protein B-cell lymphoma-2 (Bcl-2) survival ex vivo but rendered partially dispensable if mitochondrial respiration inhibited. Together demonstrate tissue-resident, population independently survive microenvironment.
