ABSTRACT
Osteoporosis, a common skeletal disorder mainly affecting postmenopausal women, is characterized by the imbalance between osteogenesis and osteoclastogenesis. Circ_0134944 has been recently found to be upregulated in osteoporosis (PMOP) patients. However, its role remains unknown. Here we aimed explore of circ_0134944 reveal underlying mechanism. qRT-PCR was used determine expression circ_0134944, miR-127-5p, PDX1 SPHK1 blood mononuclear cells (BMCs) PMOP Bone marrow mesenchymal stem (BMSCs) were as cellular model. Western blotting osteogenesis-related genes (Runx2, OPN, OCN). ALP Alizarin Red S staining performed evaluate osteogenic differentiation. The interactions miR-127-5p PDX1, determined dual-luciferase reporter ChIP assay. Circ_0134944, while downregulated Enhanced suppressed osteogenesis, which then reversed overexpression. binding confirmed Moreover, enriched promoter region SPHK1, overexpression prevented promotion induced Taken together, these results demonstrate that inhibit via miR-127-5p/PDX1/SPHK1 axis. Thus, present study offered evidence circ_0134944/miR-127-5p/PDX1/SPHK1 axis could potential therapeutic target for PMOP.
