ABSTRACT
No methods for isolating induced alveolar epithelial progenitor cells (AEPCs) from human embryonic stem (hESCs) and pluripotent (hiPSCs) have been reported.Based on a study of the stepwise induction (AECs), we identified carboxypeptidase M (CPM) as surface marker NKX2-1 + ''ventralized'' anterior foregut endoderm (VAFECs) in vitro fetal murine lungs.Using SFTPC-GFP reporter hPSCs 3D coculture system with lung fibroblasts, showed that CPM isolated VAFECs differentiate into AECs, demonstrating is AEPCs.Moreover, differentiation formed spheroids lamellar-body-like structures an increased expression surfactant proteins compared 2D differentiation.Methods to induce isolate AEPCs using consequently generate would aid pulmonary disease modeling regenerative medicine.
