ABSTRACT
OBJECTIVES/GOALS: Susceptible mucocutaneous membranes of the eye and nasal cavity are easily infected by viruses leading to pink or respiratory infections whose direct cost has been estimated as $16 billion annually in United States. We have developed a novel effective barrier that will be agnostic variants enveloped like coronaviruses. METHODS/STUDY POPULATION: evaluated efficacy hydroxypropyl cyclodextrin preventing coronavirus using 25 humanized angiotensin converting enzyme-2 receptor (hACE-2) mice under BSL3 laboratory setting. shown is safe efficacious vitro cell lines. instilled 10 uL aliquot into nostril mouse 30 minutes before exposing them 10uL titer containing 10,000 plaque forming units SARS-CoV-2 delta variant. The control received infection but not barrier. were observed for 5 days after which they sacrificed. analyzed lungs palates viral load reverse transcription-polymerase chain reaction. RESULTS/ANTICIPATED RESULTS: our variant hACE2 models. secretions treated less infectious with lower than mice. showed decrease IFN gene expression many cytokines chemokines regulate virally induced inflammatory responses such IL-1b, IL-8, CXCL9, CXCL10, CCLs. plasma Angiotensin I II decreased treatment, correlating lungs. These peptides may useful biomarkers monitoring within individuals. DISCUSSION/SIGNIFICANCE: This supports barrier’s reduce transmission prevent SARS-CoV-2. easy use can augment layers cavity. Our economic public health burden seasonal their related deaths amongst public.
