ABSTRACT
Triple-negative breast cancer (TNBC), accounting for approximately 20% of cases, is a particular subtype that lacks tumor-specific targets and difficult to treat due its high aggressiveness poor prognosis. Chemotherapy remains the major systemic treatment TNBC. However, applicability efficacy in clinic are usually concerning lack targeting, adverse side effects, occurrence multidrug resistance, suggesting development effective therapeutics still highly demanded nowadays. In this study, an injectable alginate complex hydrogel loaded with indocyanine green (ICG)-entrapped perfluorocarbon nanoemulsions (IPNEs) camptothecin (CPT)-doped chitosan nanoparticles (CCNPs), named IPECCNAHG, was developed photochemotherapy against IPNEs can induce hyperthermia generate more singlet oxygen than equal dose free ICG upon near-infrared (NIR) irradiation achieve photothermal photodynamic therapy. CCNPs positive charge may facilitate cellular internalization provide sustained release CPT carry out chemotherapy. Both nanovectors stabilize agents same system without interactions. IPECCNAHG integrating enables stage-wise combinational overcome issues described above. With 60 s NIR irradiation, significantly inhibited growth MDA-MB-231 tumors mice toxicity within 21 day treatment. We speculate such anticancer accomplished by phototherapy followed chemotherapy, where cells were first destroyed IPNE-derived oxygen, damage after CCNPs; two-stage tumoricidal process. Taken together, anticipated be feasible tool TNBC clinic.
