ABSTRACT
Nicotinamide adenine dinucleotide (NAD+) plays a vital role in cellular processes that govern human health and disease. phosphoribosyltransferase (NAMPT) is rate-limiting enzyme NAD+ biosynthesis. Thus, boosting level via an increase NAMPT levels attractive approach for countering the effects of aging metabolic This study aimed to establish IRW (Ile-Arg-Trp), small tripeptide derived from ovotransferrin, as booster levels. Treatment muscle (L6) cells with increased intracellular protein (2.2-fold, p < 0.05) boosted (p 0.01). Both immunoprecipitation recombinant assays indicated possible NAMPT-activating ability Similarly, mRNA liver (2.6-fold, 0.01) tissues (2.3-fold, C57BL/6J mice fed high-fat diet (HFD). A significantly circulating was also observed following treatment (4.7 fold, 0.0001). Dosing Drosophila melanogaster elevated both D-NAAM (fly NAMPT) vivo 0.05). However, did not boost SIRT1 KO cells, indicating dependency pharmacological effect. Overall, these data indicate novel peptide pool.
