ABSTRACT
Nutritional induction of endogenous antimicrobial peptide expression is considered a promising approach to inhibit the outgrowth and infection pathogenic microbes in mammals. The present study investigated possible regulation porcine epithelial β-defensins response branched-chain amino acids (BCAA) vivo vitro. BCAA treatment increased relative mRNA jejunal ileal weaned piglets. In IPEC-J2 cells, isoleucine, leucine, valine could stimulate β-defensin expression, possibly associated with stimulation ERK1/2 phosphorylation. Inhibition Sirt1 ERK completely blocked activation 90RSK protein by simultaneously decreasing defensin expression. intestinal isoleucine most, potent through Sirt1/ERK/90RSK signaling pathway. lipopolysaccharide was related an ERK-independent modulation might be enhance disease resistance health young animals children.
