Engineering Saccharomyces cerevisiae for Enhanced Production of Protopanaxadiol with Cofermentation of Glucose and Xylose

ABSTRACT
Protopanaxadiol (PPD), an active triterpene compound, is the precursor of high-value ginsenosides. In this study, we report a strategy for enhancement PPD production in Saccharomyces cerevisiae by cofermentation glucose and xylose. mixed sugar fermentation, strain GW6 showed higher titer yield than that obtained from cultivation. Then, engineering strategies were implemented on to enhance yields, such as adjustment central carbon metabolism, optimization mevalonate pathway, reinforcement xylose assimilation regulation cofactor balance, namely, overexpression xPK/PTA, ERG10/ERG12/ERG13, XYL1/XYL2/TAL1, POS5, respectively. particular, final GW10, harboring overexpressed exhibited highest yield, which was 2.06 mg PPD/g sugar. 5-L fermenter, reached 152.37 mg/L. These promising results demonstrate great advantages over high-yield PPD.
