ABSTRACT
Nowadays, massive genomics and transcriptomics data can be generated at the single-cell level. However, proteomics in this setting is still a big challenge. Despite great improvements sensitivity performance of mass spectrometry instruments better knowledge on sample preparation processing, it widely acknowledged that multistep workflows may lead to substantial loss, especially when working with paucicellular samples. Still, clinical fields, frequently limited amounts are available for downstream analysis, thereby hampering comprehensive characterization protein To aim peptide recoveries, we compare existing novel approaches protocols studies, from collection, cell lysis, quantification, electrophoresis/staining digestion, recovery, LC-MS/MS instruments. From critical evaluation, conclude recent innovations technologies, together high quality management samples, make samples possible, which will have immediate impact community.
