ABSTRACT
Single-molecule detection of nucleic acids in body fluids is vital but challenging. This work presents an optical microfiber biosensor with a metal-semiconductor-2D material hybrid nanointerface for single-molecule amplification-free complex fluids. By optimizing the components, we achieved significant enhancement evanescent field, enabling ultrahigh sensitivity at surface. It allowed DNA molecules level and could identify single-base-pair mismatches. Utilizing microscale diameter millimeter-length design, overcomes limitations associated nanosensors, providing practical solution point-of-care diagnostics. The sensor demonstrated its potential through ultrasensitive HIV such as serum, sweat, saliva. advancement marks critical step forward acid detection, facilitating early disease diagnosis, personalized medicine, fundamental biological research, despite challenges posed by nanosize, chain-like morphology, environmental interference acids.
