Polyaniline-Based Glyco-Condensation on Au Nanoparticles Enhances Immunotherapy in Lung Cancer

ABSTRACT
Lung cancer is considered among the deadliest cancers with a poor prognosis. Au@PG nanoparticles (NPs) are gold (Au)-based NPs featuring polyaniline-based glyco structure (PG) generated from polymerization of ortho-nitrophenyl-β-d-galactopyranoside (ONPG) promising M1 macrophage polarization activity, resulting in tumor remodeling and cold to hot microenvironment, which promotes cytotoxic T cell response inhibition. The combination anti-programmed death protein 1 (PD-1) therapy improved inhibition immunosuppression, accompanied by secretion immunogenic cytokines. A one-pot synthetic method was developed achieve glyco-condensation during formation NPs, induced more efficiently than Au@glucose, Au@mannose, Au@galactose NPs. switch M2 macrophages dependent on NP size, smaller performing better larger ones, effectiveness ranked as follows: 32.2 nm ≈ 29.8 < 26.4 18.3 nm. Cellular uptake endocytosis size-dependent endoplasmic reticulum (ER) stress, resulted activation spleen tyrosine kinase (SYK), leading immune modulations polarization. Our results suggested potential lung immunotherapy.
