Neutrophil-Biomimetic “Nanobuffer” for Remodeling the Microenvironment in the Infarct Core and Protecting Neurons in the Penumbra via Neutralization of Detrimental Factors to Treat Ischemic Stroke

ABSTRACT
High level of detrimental factors including reactive oxygen species (ROS) and inflammatory cytokines accumulated in the infarct core their erosion to salvageable penumbra are key pathological cascades ischemia-reperfusion injury stroke. Few neuroprotectants can remodel hostile microenvironment for failure interfere with dead or biofunctionally inactive dying cells. Even is temporarily attenuated by medications; insults from still erode continuously along drug metabolism clearance. Herein, a strategy named "nanobuffer" proposed neutralize buffer destructive penumbra. Inspired neutrophils' tropism affinity cytokines, poly(lactic-co-glycolic acid) (PLGA) nanoparticles coated neutrophil membrane target absorb cytokines; α-lipoic acid decorated on surface cannabidiol loaded ROS scavenging neuroprotection, respectively, construct basic unit nanobuffer. Such nanobuffer exerts comprehensive effect area via factor neutralization cannabidiol-induced neuroprotection. Besides, possibly be enhanced dynamic ROP (ring-opening-polymerization)-induced cross-fusion among closely adjacent units vivo. Systematic evaluations show significant decrease penumbra, reduced volume, improved neurological recovery compared untreated group stroke rats.
