Adhesive Ergothioneine Hyaluronate Gel Protects against Radiation Gastroenteritis by Alleviating Apoptosis, Inflammation, and Gut Microbiota Dysbiosis

ABSTRACT
Radiation gastroenteritis represents one of the most prevalent and hazardous complications abdominopelvic radiotherapy, which not only severely reduces patients' life quality but also restricts radiotherapy efficacy. However, there is currently no clinically available oral radioprotector for this threatening disease due to its complex pathogenesis harsh gastrointestinal environment. To end, study developed a facile effective radioprotector, ergothioneine hyaluronate (EGT@HA) gel, protecting against radiation by synergistically regulating oxidative stress, inflammation, gut microbiota. In vitro cellular experiments verified chemical stability free radical scavenging ability EGT favorable radioprotective efficacy inhibiting intracellular reactive species (ROS) generation, DNA damage, mitochondrial apoptosis. At in vivo level, EGT@HA with prolonged residence mitigated radiation-induced tissue injury, apoptosis, neutrophil infiltration, flora dysbiosis. For first time, work investigated protective effects gel on gastroenteritis, hastens advancement novel provides valuable radioprotection paradigm modulating microbiota disturbance.
