ABSTRACT
Circulating extracellular vesicle (EV) PD-L1 is correlated with the occurrence and progression of lung cancer has great potential as a valuable diagnostic immunotherapy predictive biomarker. In this work, we propose fluorescent biosensing method for sensitive accurate determination circulating EV PD-L1. Specifically, after phosphatidylserine-targeting peptide-assisted magnetic enrichment, programmable DNA circuit designed to translate presence appearance numerous duplex probes on surface. Upon fructose treatment, these newly formed are released from surface activate trans-cleavage activity CRISPR/Cas12a system, which finally produces significant fluorescence signal. Experimental results reveal that not only enables detection limit 67 particles/mL but also demonstrates use in diagnosis response prediction principle-of-proof study. Therefore, may provide useful tool determination, information precise personalized treatment patients.
