ABSTRACT
Reactive oxygen species (ROS)-involved photodynamic therapy (PDT) and chemodynamic (CDT) hold great promise for tumor treatment. However, hypoxia, insufficient H2O2, overexpressed glutathione (GSH) in the microenvironment (TME) hinder ROS generation significantly. Herein, we reported CaO2@Cu-TCPP/CUR with O2/H2O2/Ca2+ self-supply GSH depletion enhanced PDT/CDT Ca2+ overload synergistic therapy. CaO2 nanospheres were first prepared used as templates guiding coordination between carboxyl of tetra-(4-carboxyphenyl)porphine (TCPP) Cu2+ ions hollow CaO2@Cu-TCPP, which facilitated GSH-activated TCPP-based PDT Cu+-mediated CDT. The structure was then loaded curcumin (CUR) to form composites. Cu-TCPP prevented degradation CaO2, while reacted deplete GSH, produce Cu+ ions, release TCPP, CUR. H2O generate O2, achieve PDT, CDT, CUR-enhanced Thus, this multilevel amplifier enhances fewer side effects drug resistance.
