ABSTRACT
An integrated distance-based origami paper analytical device (ID-oPAD) is developed for simple, user friendly and visual detection of targets interest. The platform enables complete integration target recognition, signal amplification, output based on aptamer/invertase-functionalized sepharose beads, cascaded enzymatic reactions, a 3D microfluidic paper-based with readout, respectively. invertase-DNA conjugate released upon addition, after which it permeates through the cellulose flows down into bottom zone, whereas beads larger size are excluded stay in upper zone. Finally, initiates reactions translates brown bar chart reading. By simply closing device, ID-oPAD sample-in-answer-out assay within 30 min quantitative readout. Importantly, bound/free probe separation achieved by taking advantage difference between pores, downstream amplification realized compatibility multiple enzymes corresponding substrates. Overall, advantages low-cost, disposability, simple operation, offers an ideal point-of-care testing, especially resource-limited areas.
