ABSTRACT
Circulating tumor cells (CTCs) in blood is the direct cause of metastasis. The isolation and detection CTCs whole very important clinical value early diagnosis, postoperative review, personalized treatment. It difficult to separate all types that efficiently rely on a single path due cancer cell heterogenicity. Here, we designed new kind "filter chip" for retention with high efficiency by integrating effects size specific antigens surface cells. filter chip consists semicircle arc arrays can large-scale CTC microspheres, which combined automatically. We synthesized interfacial zinc oxide coating nanostructure microsphere increase area enhance capturing CTCs. Microspheres, trapped arrays, would entrap CTCs, too. combination three kinds strategies resulted more than 90% capture different lines. Furthermore, it easy find isolate circulating from as be fixed inside structure chip. To avoid background contamination when few are surrounded millions nontarget cells, digital method was applied improve sensitivity. were specifically labeled antibody–DNA conjugates detected via DNA signal amplification. strategy antibody–functional microsphere-integrated microchip sorting may broad implications favor fundamental biology research, precise monitoring clinics.
