ABSTRACT
The exosome is a promising biomarker carrying many kinds of membrane proteins with huge heterogeneity, so the sensitive and multiplex analysis exosomes very significant for disease diagnosis exploration their biological functions. Herein, we propose an efficient method highly detection heterogeneity identification based on design fabrication aptamer-coated liposome complex coupled terminal deoxynucleotidyl transferase (TdT)-mediated polymerization. Specifically, in presence target exosomes, aptamers immobilized surface 1,2-dioleoyl-3-trimethylammonium-propane liposomes prefer to bind exosomal due high affinity. resulting aptamer–exosome will be accessible TdT switch polymerization reaction signal amplification, achieving exosomes. Furthermore, proposed can employed profile different by making use cluster corresponding obtain fingerprint map various cancer cell-derived Thus, our approach may provide robust strategy advantages being label-free having no separation, potentially enabling precise subpopulation practical value clinical applications.
