ABSTRACT
Efficient methods for the site-specific installation of structurally defined interstrand cross-links in duplex DNA may be useful a wide variety fields. The work described here developed high-yield synthesis chemically stable resulting from reductive amination reaction between an abasic site and noncanonical nucleobase 2-aminopurine DNA. Results footprinting, liquid chromatography-mass spectrometry, stability studies support formation N2-alkylamine attachment residue Ap site. performs best when on opposing strand is offset 1 nt to 5'-side cross-link confers substantial resistance thermal denaturation (melting). cross-linking fast (complete 4 h), employs only commercially available reagents, can used generate cross-linked duplexes sufficient quantities biophysical, structural, repair studies.
