ABSTRACT
Rapid and accurate diagnosis of cardiovascular diseases (CVDs) at the earliest stage is paramount importance to improve treatment outcomes avoid irreversible damage a patient's system. Microfluidic paper-based devices (μPADs) represent promising platform for rapid CVD point care (POC). This paper presents an electrochemical μPAD (E-μPAD) with all-in-one origami design POC testing cardiac protein markers in whole blood. Based on label-free, impedance spectroscopy (EIS) immunoassay, E-μPAD integrates all essential components single chip, including three cells, plasma separation membrane, buffer absorption pad, enabling easy streamlined operations multiplexed detection [cardiac troponin I (cTnI), brain natriuretic peptide (BNP)-32, D-Dimer] finger-prick blood sample within 46 min. Superior analytical performance achieved through sensitive EIS measurement carbon electrodes decorated semiconductor zinc oxide nanowires (ZnO NWs). Using spiked human samples, ultralow limits (LODs) are 4.6 pg/mL (190 fM) cTnI, 1.2 (40 BNP-32, 146 (730 D-Dimer. Real samples purified proteins also tested, device's was proven be comparable commercial ELISA kits. The will allow operations, which holds great potential on-site screening acute CVDs nonlaboratory settings such as emergency rooms, doctor's offices, or patient homes.
