Engineered Spore-Forming Bacillus as a Microbial Vessel for Long-Term DNA Data Storage

ABSTRACT
DNA data storage technology may supersede conventional chip or magnetic medium, providing long-term stability, high density, and sustainable storage. Due to its error-correcting capability, stored in living organisms exhibits fidelity information replication. Here we report the development of a Bacillus chassis integrated with an inducible artificially assembled bacterial chromosome facilitate random access. We generated three sets form sequences using rudimentary coding system accessible by regulatory promoter. Sporulated harboring genes were used for storage, where viability assays spores subjected harsh environmental stresses evaluate stability. The accuracy remained above 99% after temperature oxidative stress treatment, whereas UV irradiation treatment provided 96% accuracy. developed artificial larger datum volume other digital encoding decoding programs.
