Gold nanoclusters-assisted delivery of NGF siRNA for effective treatment of pancreatic cancer

ABSTRACT
Abstract Pancreatic cancer is one of the deadliest human cancers, whose progression highly dependent on nervous microenvironment. The suppression gene expression nerve growth factor (NGF) may have great potential in pancreatic treatment. Here we show that gold nanocluster-assisted delivery siRNA NGF (GNC–siRNA) allows efficient silencing and GNC–siRNA complex increases stability serum, prolongs circulation lifetime blood enhances cellular uptake tumour accumulation siRNA. potently downregulates Panc-1 cells tumours, effectively inhibits three models (subcutaneous model, orthotopic model patient-derived xenograft model) without adverse effects. Our study constitutes a straightforward but effective approach to inhibit via knockdown, suggesting promising therapeutic direction for cancer.
