ABSTRACT
The mitochondrial unfolded protein response (UPRmt) is a mitochondrial-to-nuclear signaling pathway that activated to maintain function when there an accumulation of misfolded proteins within mitochondria. Mitochondrial essential for chondrocyte homeostasis, and dysfunction characteristic osteoarthritis (OA). However, the role UPRmt in OA remains unclear. In present study, level was examined primary mouse chondrocytes subjected different stresses articular cartilage model mice patients. relationship between activation progression studied. induced diverse mice. Enhancement with nicotinamide riboside (NR) significantly improved function, reduced death, attenuated pain, ameliorated progression, protective effects decreased chondrocyte-specific Atf5 knockout (ATF5f/fCol2a1-CreERT2) induction also identified patients associated less severe hip lower levels inflammation synovial fluid. These findings identify exposed pathological ameliorates suggesting exerts effect against may be potential diagnostic therapeutic strategy OA.
