ABSTRACT
Abstract Photodynamic therapy is an effective alternative to traditional treatments due its minimally invasive nature, negligible systemic toxicity, fewer side effects, and avoidance of drug resistance. However, it still challenging design photosensitizers with high singlet oxygen ( 1 O 2 ) quantum yields (QY) severe aggregation the hydrophobic photosensitizers. Herein, we developed a discrete organoplatinum(II) metallacage using therapeutic cis -(PEt 3 Pt(OTf) as building block improve QY, thus achieving synergistic anticancer efficacy. The metallacage-loaded nanoparticles (MNPs) tri-modality imaging capability allow precise diagnosis tumor real-time monitoring delivery, biodistribution, excretion MNPs. MNPs exhibited excellent anti-metastatic effect superior anti-tumor performance against U87MG, resistant A2780CIS, orthotopic models, ablating tumors without recurrence after single treatment. Gene chip analyses confirmed contribution different modalities abrogation. This supramolecular platform holds potential in cancer theranostics.
