ABSTRACT
Abstract Recent years have seen an increase of extracellular vesicle (EV) research geared towards biological understanding, diagnostics and therapy. However, EV data interpretation remains challenging owing to complexity biofluids technical variation introduced during sample preparation analysis. To understand mitigate these limitations, we generated trackable recombinant (rEV) as a reference material. Employing complementary characterization methods, demonstrate that rEV are stable bear physical biochemical traits characteristic EV. Furthermore, can be quantified using fluorescence-, RNA- protein-based technologies available in routine laboratories. Spiking allows recovery efficiencies commonly implemented separation methods identified, intra-method inter-user variability induced by handling defined, normalize improve sensitivity enumerations. We anticipate will aid EV-based analysis, normalization, method development instrument calibration various biomedical applications.
