ABSTRACT
Abstract Drug-induced cardiotoxicity arises primarily when a compound alters the electrophysiological properties of cardiomyocytes. Features intracellular action potentials (iAPs) are powerful biomarkers that predict proarrhythmic risks. In last decade, number vertical nanoelectrodes have been demonstrated to achieve parallel and minimally-invasive iAP recordings. However, large variability in success rate signal strength hindered from being broadly adopted for proarrhythmia drug assessment. this work, we develop vertically-aligned nanocrown electrodes mechanically robust > 99% rates obtaining access through electroporation. We validate accuracy electrode recordings by simultaneous patch clamp recording same cell. Finally, demonstrate enable prolonged continual monitoring cells upon sequential addition four incremental doses. Our technology development provides an advancement towards establishing screening assay preclinical evaluation drug-induced arrhythmogenicity.
