Integrative multi-region molecular profiling of primary prostate cancer in men with synchronous lymph node metastasis

ABSTRACT
Abstract Localized prostate cancer is frequently composed of multiple spatially distinct tumors with significant inter- and intra-tumoral molecular heterogeneity. This genomic diversity gives rise to many competing clones that may drive the biological trajectory disease. Previous large-scale sequencing efforts have focused on evolutionary process in metastatic cancer, revealing a potential clonal progression castration resistance. However, origin synchronous lymph node (LN) metastases primary disease still unknown. Here, we perform multi-region, targeted next generation construct phylogenetic trees men LN metastasis better define pathologic features most likely spread LNs. Collectively, demonstrate combination histopathologic factors, including tumor grade, presence extra-prostatic extension, cellular morphology, oncogenic alterations are associated metastasis.
