ABSTRACT
Abstract Somatic mutations in Mediator complex subunit 12 (MED12m) have been reported as a biomarker of uterine fibroids (UFs). However, the role MED12m is still unclear pathogenesis UFs. Therefore, we investigated differences DNA methylome, transcriptome, and histological features between MED12m-positive -negative methylomes transcriptomes were obtained from UFs myometrium, hierarchically clustered. Differentially expressed genes comparison with myometrium co-expressed detected by weighted gene co-expression network analysis subjected to ontology enrichment analyses. The amounts collagen fibers number blood vessels smooth muscle cells histologically evaluated. Hierarchical clustering based on methylation clearly separated MED12m-positive, MED12m-negative had increased activities extracellular matrix formation, whereas angiogenic cell proliferation. different methylation, expression, features. form tumor rich poor compared UFs, suggesting MED12 affect tissue composition
