Induction of autophagy improves embryo viability in cloned mouse embryos

ABSTRACT
Abstract Autophagy is an essential cellular mechanism that degrades cytoplasmic proteins and organelles to recycle their components. Moreover, autophagy for preimplantation development in mammals. Here we show also important reprogramming somatic cell nuclear transfer (SCNT). Our data indicate unlike fertilized oocytes, not triggered SCNT embryos during 6 hours of activation. Mechanistically, the inhibited autophagic induction activation due cytochalasin B (CB) caused depolymerization actin filaments. In this study, induced by rapamycin pp242, which could restore expected level significantly enhance blastocyst stage when compared with control (68.5% 68.7% vs. 41.5%, P < 0.05). Furthermore, treatment pp242 accelerates active DNA demethylation indicated conversion 5 mC hmC improves degradation maternal mRNA as well. Thus, our findings reveal might be one serious causes low efficiency SCNT.
