ABSTRACT
Abstract It is known that oxidative stress plays a pivotal role in age-related macular degeneration (AMD) pathogenesis. Alpha-mangostin the main xanthone purified from mangosteen as anti-oxidative properties. The aim of study was to test protective effect alpha-mangostin against both retina light-damaged mice model and hydrogen peroxide (H 2 O )-stressed RPE cells. We observed significantly inhibited light-induced photoreceptors 200 μM H -induced apoptosis generation reactive oxygen species (ROS) malondialdehyde (MDA) were suppressed by alpha-mangostin. stimulation resulted an increase superoxide dismutase (SOD) activity, glutathione peroxidase (GPX) activity (GSH) content vivo vitro. Furthermore, mechanism retinal protection involves accumulation nuclear translocation NF-E2-related factor (Nrf2) along with up-regulation expression heme oxygenas-1 (HO-1). Meanwhile, can activate PKC-δ down-regulate mitogen-activated protein kinases (MAPKs), including ERK1/2, JNK, P38. results suggest could be new approach suspend onset development AMD.
